Membrane chromatography technologies for rapid mRNA purification
The development of messenger RNA (mRNA)-based therapeutics has grown significantly in recent years, with the potential to revolutionize the treatment of such diseases and disorders as cancers, genetic disorders, and infectious diseases. mRNA production starts with supercoiled plasmid DNA (SC pDNA), which is linearized for use in an in vitro transcription reaction to generate the mRNA product. A critical final step in mRNA production is the purification process, which directly impacts the quality, efficacy, and safety of the ultimate product. With increasing demands for personalized mRNA medicine and larger-size mRNA forms (such as self-amplifying mRNA), novel purification tools are needed to reduce purification times and enhance productivity.
In this presentation, Purilogics by Donaldson introduces the concept of using high-performance membrane chromatography technologies to rapidly purify SC pDNA and mRNA. We will illustrate purifying SC pDNA by capture chromatography using Purexa™ NAEX Plus, a weak anion chromatography membrane, followed by a polishing step using Purexa™ MCP, a hydrophobic interaction chromatography membrane that has superior and consistent capability to separate SC pDNA from open circular pDNA. We will illustrate purifying mRNA using Purexa™ OdT, an affinity capture chromatography membrane targeting the Poly A tails in mRNA.
The Purexa™ purification products are pre-packed, ready-to-use chromatography devices designed for user-friendly operation with short residence times. Suited for different scales, these products can be used as single-use devices or in multiple cycles, providing adaptability to various processes and meeting diverse requirements.

Jinxiang (Jin) is an inventor and co-founder of Purilogics. Jin is a graduate from National Science Foundation Innovation-Corps program, and he is an internationally recognized expert in the field of polymeric thin-film composite membranes.
Jin has spent 10+ years developing high-performance membranes and processes for chemical and biologic purification. Jin's leadership has included service as Vice President at Purilogics managing the company’s day-to-day operation, and Principal Investigator on multiple Purilogics small business innovation research (SBIR) grants to build high capacity and high selectivity membrane chromatography products for biologic purification. Jin earned PhD in Chemical Engineering from Clemson University.
